Concise is the problem ;-) It is a 20 year story with several false starts and dead ends. I'd be happy to email James Madigan the introduction to my thesis as one place to start, since it goes through the basics of the story. For example, Ho found that HIV was replicating rapidly by measuring the rate of virus RNA decline (by PCR) once antivirals were initiated.
